Just a few things...

1) Go into the control panel (right click on icon in tray and select
control panel). On the diagnostics tab there is information that is
testing your port connections, etc. (9000, 3483). Do these all say OK?

2) Have you changed the name or path of your music
directory/directories?

3) Are you starting LMS as Administrator?

If everything above checks out, I would uninstall LMS and reinstall.
