I have the same problem with my FT290RII and the RS232.
Interface WM Nomic feed by the mic/speaker jacks of my laptop. RC1 works fine. I do run this as a second instance but even if I close the first instance for my 7300 and restart the 290 one it still does it.
Windows 7
Rig none
RS232 com 1 RTS for PTT no CAT control
Mode none
Split none
It keys radio but no audio and some times doesn't key the radio. This happens with Enable or the Tune buttons If I stop it and try again it keys and has audio. It may work a couple times then either no audio or no key. However WSJT acts like it is working. If you need further information please let me know. If I do not answer this evening I will tommorow morning as I get up at 3AM EDT.

I see exactly the same as you with RS232 PTT.
I hadn't monitored the state of the Test PTT button. As you observe, if this turns red during a receive period then the following transmission has no audio. If the button does not turn red during a receive period the following transmission is OK. Watched the behaviour for a number of periods and this seems to be consistent.

To start with, my system is a Dell 1650, i7 @3.40 GHz, 16GB RAM, W10 pro 64bit. I do not use CAT but instead use RTS on COM1 for opto-isolated PTT and XFR isolation for audio in and out of the sound-card, and have done so without issue since 2006.
Two problems : -
1. I have found with rc2 that occasionally PTT is not working at all and when it does there is often no TX audio present either. While monitoring 'Radio' under 'Settings' I have noticed when this occurs, after a successful transmission about two seconds into the RX period the 'Test PTT' button turns red and from that point there is no PTT in the following TX period. After a time it appears to clear itself and PTT in the following period with or without TX audio.
2. I am still getting numerous false decodes. Most of these contain '/R' and '<.....>'. I am not using AP.
