True, the one dot is in a different location but with no other letters or numbers to guide your finger, it would make reading it difficult. I did a set of lottery cards with brail numbers and many people got some numbers confused when read up-side-down. So I put three dots under the numbers and removed the leading zero in one digit numbers Since I was using half inch square dymo I put the three dots in the cell below the number to give more air around the dots in the number.

And, if you really wanted to label a two digit die, such as a d20, you
could put on both digits, which you pretty much have to do anyway, so
1 becomes 01, 10 is 10.

The most dots in that system is seven, on a 07. The least is 11, with
two. If you just distributed the weight of the dots, you could ensure
an even more random roll, though the difference of five braille dots
is relatively negligible.
