[Histonet] 5 Yr old Leica ST5020 is down
Hello Histology friends, We are a research lab with a 5 year old Leica ST5020 autostainer that looks like new. We run less than 5000 slides a year on it. We noticed that (of course just after the warranty ended) that one of the water rinse stations stayed full of water all the time, even when the instrument was off. It may have been like this from the beginning and we just did not notice it. Leica quoted a $6000 repair but said we could run it as is. So, we skipped the repair and went on using it. Now the instrument is completely down and the repair is a new "slave controller board" for $17,600, stepper motor for arm, etc. with total quote for over $23,800. Has anyone else had major issues with this instrument over time. We are trying to decide whether to fix it or go for a new stainer. I'm kinda wondering if we got a lemon. Hello Dr. Rosen and Histonetters, We have used a Leica Bond RX for over 2 years now, cleaning and caring for it just as you do. My work is mostly animal which is why we mainly choose the red chromogen alkaline phosphatase detection kit. We see the same artifact sporadically. Sometimes we will see it for several days and then maybe a week goes by and we don't see any of it. It may only be a small amount and I have seen it basically all over the slide. I know it is not related to our tissues or slides. Our Leica rep came out and increased washes but it really doesn't help. If anyone has a solution to the problem, I would also be interested. The artifact is usually golden brown to red/pink in color, is clustered, appears both on top of the tissue and around it. If we do an aggressive rundown, it appears less red/brown and bluer. We are using a Leica Bond MAX IHC system which is about a year old. We usually do two runs a day and 95% of the time we only use a red detection kit. We only see this on our red detection kit on our Leica bond. The brown is unaffected. The corresponding H slides are fine. We started having this artifact after a month or two of production. 85% of the time we have some sort of artifact. Sometimes its perfect, sometimes it makes the tissue unreadable. We haven?t figured out any precipitating factors. We have tried a number of things, but we still can?t get control of this. Here is a brief overview of our protocols: - Processor: Leica ASP300s tissue processor. -Parrafin = Mercedes HWWX (dual purpose embedding and infiltration parrafin) melting point 57c ? we tried switching, and it didn?t help. - We only use distilled water in our baths - Mercedes Starfrost charged slides - Oven for 30 min at 62c - We always use a cold red detection kit -Sometimes our runs are delayed but not more than 4 hours - Rundown is done in our stainer (standardized), and slides are coverslipped with tape. - The bulk fluid containers in the bond are cleaned weekly and refrigerated between runs. - The probe is cleaned every 300 slides. - Mixing wells are cleaned out every two weeks and replaced often. -Covertiles are well tended to. We have tried to correct this problem, but we can't seem to fix it. I should note that cutting at 2 isn't required, but it is desired once they saw that I can. Rhesus is a species of old world monkey. There are a couple of other companies that have this same clone and on their datasheets they say it cross reacts with rhesus monkey. The CD34s (2) that I have do not state on their data sheets that they cross react with anything except human and yet are the same clone as the companies that say it does. I guess I will just break down and purchase a different clone that states it works in rhesus monkey. 2 more bits of information about my dilemma; I did try staining without any retrieval - no signal I am using rhesus kidney as a control (getting no signal), but I also ran some human kidney and got beautiful staining with a 20 minute HIER using a pH 8 EDTA buffer. So I know the antibodies and my protocols work, just not on my rhesus. Does anyone know of a CD34 that will work in this species? More suggestions please? Thank you all for your time. Good morning my fellow Histo-netters, Does anyone have experience with any CD34 antibody that would work in FFPE rhesus tissues? A few companies have one with clone QEBend 10 and say that it works but I have had no luck. I am using the Bond RX polymer system and I've tried all the epitope retrievals from Leica. We have upgraded to a Dako Link 48 machine and have a working Dako Autostainer system to get rid of. The machine was purchased new in 2000 and has been well maintained. The working computer and printer are included, as well as jugs, manual, and software backups. For the first time my pathologist would like to bring an ASR antibody in house. It is made by Biocare, and our validation slides look good compared to the other clone we want to get rid of. However, I see in question 12425 of my checklist that I have to establish or verify the performance characteristics... in accordance with the Method Performance Specifications section of the All Common Checklist. [Histonet] lower glands in colon not staining with IHC
On our AE1/AE3 and CK 20 slides we are noticing a deminished staining in the lower glands of the colon sections. My first thought was inadequate fixation, however the BerEP4 stained evenly. This has been happening on different tissues, including small biopsies. Does anyone have any ideas as to something else that could be causing a lack of staining on the internal colon portions? We use Dako reagents and polymer kit and the skin and other tissues in the sausage block stain beautifully. We are using a similar method at our site. The aspirate is placed into EDTA and then into formalin to fix. We pour the aspirate through a biopsy bag, open the bag and scrape the spicules together, transfer them to another biopsy bag and process as usual. If I remember correctly, the pathologist or oncologist would collect the aspirate using a heparin syringe. Some how, the aspirate smears for Hematology were made on 22x22 cover slips instead of slides. Then the EDTA tube was then given to Histology. I can't remember if we added formalin or B5 fixative to it at that time, but it was filtered into an obex tea bag type filter paper (The blood never clotted and ran through the filter paper, leaving only bone marrow spicules in the paper). We would fold the paper, place it in the cassette, and process as usual. The pathologist preferred this method because he wouldn't have to look at levels of clot to find spicules. We use Bond slides and have noticed that occasionally they seem to have an extra thick coating on them that will pick up DAB and turn slides brown. If the glass itself is turning brown this might be your issue. Have you tried different slide lots or brands? [Histonet] Sakura glas coverslipper and mounting media types
For those with the Sakura Glas coverslippers, what type of mounting media are you using in these? Sakura recommends only their brand of course, but I want something that dries faster. We do not have excess mounting media on the outside of our slides, but after being in the oven for three days we had several slides sticking. It looked like the mounting media slipped down the slide to pool at the bottom and stick. I am thinking their media is too viscous. If I turn the amount down on the machine any further we end up with not enough media on the slides and get large air pockets after just a few days of storage. BTW the oven is at 40 degrees and isn't boiling hot to dry the slides. I am wondering how other hospital connected histology labs handle requests from patients to get their tissue back. I am especially interested in the placentas. [Histonet] RE: Interview Questions
The last two times we interviewed a person we gave them a written skills test before they sat down for the face to face interview. It had simple questions on it, like how to mix a percent solution, clean a formalin spill, which chemical was more hazardous than another, how to handle certain types of tissues, how to work computer applications, etc. The test has 15 questions and has a section that includes them writing on a cassette and following written instructions on how to prep a non-gyn cytology. This gave us a lot of skills information about the person and helped us weed out who was qualified. You could tell quite a bit about their knowledge just from the written portion and people seemed less nervous when there wasn't a person hovering over them. Our hospital sees this skills test as the equivalent of a typing test for transcriptionists. We add 1.0 mls of 10% aqueous Eosin to the first 100% Ethanol; we will then have Eosin in all of the Ethanol containers after they have been rotated. This removed the time consuming task of dotting each tissue at grossing and allows for easy visualization at embedding and sectioning. I have not had a complaint about the eosin interfering with FISH from our referral lab. We are using the Sakura Prisma with attached coverslipper; it is a workhorse and allows for the use of what ever reagents/stains that you prefer. We are running our HE and H Pylori stains simultaneously. We experienced a small problem with the leveling of the instrument at the initial set up but once that was corrected it has been working well since then. The only draw back that I can see with the attached coverslipper is that the slides take longer to dry than with the older model. The slides are kept horizontal rather than vertical so the xylene doesn't run off. We are not open on weekends and worked out a deal with our core l= ab that is open 24/7. When we have a breast on the processer we willrun a Sat. program that ends at 21:00. They added a task on their dai= ly checklist to come up and take the tissues off at that time. They j= ust sit them on the counter and they cool - just like they were being embed= ded. When we come in on Monday we just sit them in the embedding cent= er for 15 minutes and they warm up enough to start embedding. No harm= to tissues and no one has to come in. RE: [Histonet] problems with staining IHC
We and several other sites (in British Columbia and Alberta) that I have contacted have been experiencing IHC staining problems. Ours started in August while others have been seeing this since May. We have had conference calls with Ventana about this and they have said that it is a problem with the SuperFrost Plus slides; the slides seem to be hydrophobic. The staining that we are seeing is negative controls/positive patient, positive control/negative patient as well as variable counterstain. We have control tissue on every slide. From what we have learned from Fisher and Ventana they are working together with Erie Scientific (manufacturer of the Plus slides) to find out what is causing the problem. The slides appear to be vary from box to box and within each box. Some lot numbers are worse than others; our most recent lot number was 062510-9. I would suggest that you contact your sale person as well as the platform vendor (if your staining is automated) so they are aware of how large the problem may be. We have not found another slide that is any better than the Plus. Ventana is working on an official communication for their clients. We are very frustrated with the lack of progress in solving the problem. As anyone noticed that the Plus slides don't seem to have the same adhesive properties that they used to? [Histonet] xylene in an unventilated area
Hello all. Cytology was told that the xylene substitute the= y were using for their Surepath paps was causing staining/artifact issues. nbsp; They want to switch to using xylene in their area, but it is unventil= ated. Because of various reasons, their process can not be moved.nbs= p; They are asking me for advice about what to do for fume control. I= was wondering if a good downdraft unit would be enough to handle the xylen= e fumes. They will have maybe 400 mL of xylene uncovered while covers= lipping their paps. Does anyone else have a similar set up? The= y share their area with micro and core lab and others are complaining about= fumes even though they have borrowed a mini-hood from us. Hi all, I have ran into a troubleshooting dilemma and was wonderi= ng if anyone else could help. Is there anyone out there that uses the Biocare Medical P63 concentrated antibody with the Dako TRS and Flex detec= tion kit? I am curious what dilution you use and if you have to do an= ything special for the P63 (extra long retrieval, DAB enhancer, etc). = I am having problems getting a strong signal for the basal cells in prosta= te. I am running it at 1:50 with 40 minutes in pH 9 retrieval (which = seems too long to me but does help). A dilution with the same protoco= l at 1:100 is way too light. Hi Claire, We experienced problems with our IHC staining the fall of 2008 and it was traced back to a problem with particular lot numbers of the Superfrost slides. It was difficult to track down due to the inconsistency of the problem and the variation in the problem. We finally spoke with our Ventana technical specialist and she was able to tie our problem to problems that other facilities were experiencing. You could try contacting your Ventana technical resource person; they may be able to help. One thing we found out was that the Superfrost slides are manufactured at one site and distributed to vendors under their own name. It would be good to hear any responses to this question. Last October our entire facility's computer system was down for 2 1/2 days. We do have downtime log sheets that we implemented but no formal procedure. Management is now looking at how the crash was handled and for ways to improve things. The downtime log sheets were very useful but I'm sure there is much more that we could be doing. Have you had any problems with the Coverslipper? We purchased the Prisma with linked Coverslipper last March and have had on going problems with the Coverslipper jamming with error code -Motor ejector error E15. We have contacted Somagen and were told that this is normal. It is happening at least once a day and sometimes more. We love the stainer and if the Coverslipper wasn't jamming we would love it just as much. What do you do with the coverslipped slides? We find that they don't dry as quickly as they did on the older film Coverslipper. We don't have a vented sign out area so they are staying on the carousel until they are mostly dry. [Histonet] double immunostaining with antigen retrieval and no antigen retrieval
Dear All, Is it possible to do double immunostaining for one antibody which doesn't require antigen retrieval, and the other one which doesn't need it. Can you do the one without retrieval first, then retrieve and do the other one, or does the retrieval damage the first staining? We know that doing antigen retrieval for antibody that doesn't require it gives us non specific staining. I would appreciate some advice. Thank you very much. We were having a similar issue with our IHC staining on the Ventana Benchmark Classic. We were able to track the problem down to the slides. We never found out what was wrong with the slides but as soon as we changed lot numbers the staining issue cleared up. We were able to track down the problem with the assistance of our Ventana technical rep. Through her we were able to find out that several institutions across Canada were experiencing similar staining problems. We were all using the same lot number of slides. She also informed me that there was one manufacturer of the plus slides and they were sold under multiple vendors. Good luck with tracking down your problem. [Histonet] breaking glass jar/vials for MMA embedded specimens
Yes, the scintillation vials are notorious for shattering in the freezer. However, if you put them in a plastic container with a lid on, the shattered glass will be contained and not all over your freezer. When breaking larger glass containers, again place them in the freezer, remove from the freezer after 20 minutes or so, take the lid off, wrap jar in paper towels and hold the ends closed. Wear glovesand eye protection in case shards of glass escape. Strike the jar with a hammer and unroll the paper towel over a garbage can. Rinse the block in tap water to remove any small shards of glass. This is a great way to vent frustration!! Cathy A. [Histonet] Sanderson's Rapid Bone Stain
Dear Fellow Histonetters, There are 3 counterstains that can be used with the Sanderson's Rapid Bone Stain (RBS). We primarily used the acidified acid fuchsin, however, we had clients that only used a van Gieson counterstain. Light green can also be used. We normally stained for 1 minute ground sections in the RBS, rinsed in water and then counterstained. As Gayle mentioned, intensity of the RBS or the counterstain can be increased with addtional time. [Histonet] uncertified histotechs
wow! I've been sitting here reading these opinions and had to get my 35 years worth. I was a histotech long before I was a certified histotech. After 25 years of doing histology, I decided to get my certification for my own gratification, however in doing so it also made quit a difference in my pay scale but did not make me a better histotech as I feel my experience in the field was worth more than that piece of paper. I will put my slides up against anyone else, the ones before my certification and after. I have in the last 2 years had a cytotech who I have crosstrained in histology and she can put out a pretty good slide also. I do not know if she will take the certification or not, but she can work in my lab anytime. I have also in the last 2 years started doing IHC's. I hope to take my certification in IHC sometime in the next 25 years, however, my pathologists are quite pleased with my work. [Histonet] using patient blocks for positive IHC controls
Does anyone know if there are any regulations on how much tissue you can cut from a patient block? We currently use known positive patient slides for our IHC positive control slides? One of our pathologist questioned if we could use an entire patient block ,cutting through the block for controls as long as the slide was still positive. Or is this an individual institution decision?
