Hello all,
I was wondering if anyone has any experience with doing a trichrome and leaving
it to mordant in Bouin's for longer than 24 hours. Does this overstain and ruin
the stain?

Please clarify if you are referring to the Aniline blue, or the Hematoxylin
blue. I, also, have never heard of using Gills in association with any
Trichrome stain, but as a long time user and educator of using a one-step

IF you are not successful when the slides are stained on the Nexus OR
manually, while the controls stain well, you can eliminate the instrument as
the cause.

Sounds like you may have a cracked vortex mixer and/or silver in your reagent

You can do H&E and VVG quite nicely on this type of MMA embedded
tissue. Maybe you could also try a Sanderson's (methylene blue) with
the Van Gieson (acid fuchsin w/ picric acid) counterstain???

I am having problems with both the Goldners and Massons Trichrome on my MMA
sections. They are ground to between 20 and 30 microns, but the staining
is nonspecific. It stains some of the bone the correct color, but not the
rest of it. Any and all help would be greatly appreciated.
