yikes. I've been in the same terrible position where the staff that I support (including president, ceo, etc) are using the technology in a way that is not a good practice. the technical people (us on this list) obviously can't provide much help, because the problem is not solvable by throwing more technology at it. even if you did solve it temporarily, this bad practice would eventually lead to the same place. but, in a way, you are actually lucky because the users are obviously suffering (5 minute load time) due to the huge size of the mailbox. so, I think you can somewhat put the ball back in their court and tell them that their email will always be slow until they clean out their inbox. they have total control over the speed of their load-time.
