YES. Made exactly this last month. Copying the 400+ Mailboxes took
aprox. 20 Minutes.
This is how I did it:
1) Copy all Mailboxes and Registry to the new machine, tested the
"new" server with a test domain, Stopped pop3 and webmail on the old
server so that only new mail can change the mailboxes
2) Made an secondary MX for the new Server, changed DNS data for the
POP3&SMTP Client access to the new maschine
3) Shut down both servers completely
4) Copy only the changed domains/Users - which was very quick most
mailboxes where not affected.
5) switch on New Server - Mails started after very short time..
6) Remove the MX for the old server

I think no mail was lost. When a mailserver can't connect to his
target, he tries the secondary MX. If this als fails, he will
reque the mail and try it some times (half an hour or so) later.

No Mailserver will dump a mail after one or two wrong atempts -
Normaly this is only done after 2-3 Days.
