I have it set to open a specific file. That isn't the problem.

It is when I try to open a new file, I find that the folder opened isn't the
one I had in my default locations. I then have to open Options/Customise and
change it back again.

Check your options...settings

I you have it set to go to last used file..it will,  regardless how you got
it to open

Does anyone else find that the Default Location for files (in
Options/Customise) doesn't always "Stick".

I have two separate places where I store Legacy databases.
- one for my family files and trees that have been sent to me as Gedcoms
- one for clients' data.

I have the default location set to f:\legacydata so that I see all my own
files. But occasionally, after looking at files in the other folder
c:\jegr\legacyfiles, I find that the default location has changed to the
"clients' folder".
Sometimes I have opened the files from File/Open and other times by using
Windows Explorer and double clicking on the .fdb file. But the next time I
open Legacy and do a file open, I find it pointing at the wrong set of
files.
