Are they refusing to work on one file or all files? Try them on an RGB file
created in Photoshop. If they work on a new file, the problem lies with the
file you are working on. If that doesn't cure the problem, try resetting the
tools by Right-clicking on their icons in the Options bar.

You can also try shutting down everything running in the background by using
the three finger salute (I assume you know how to do that). That way, you
can be more sure that the problem lies with Photoshop and not another
application or utility causing a conflict.
