longer than two days do not rehydrate well. Cells appear pink with large
air-dried, pale staining nuclei - lacking chromatin details.

No the virus is the same for IPX purposes.

a little
pink still remains in the collagen since the acidic blue or green
solution will continue to differentiate the red muscle stain.
The blue collagen solution stains faster than the green solutions so use
for a shorter time.

I would dilute it to a working solution and use that.

I am not sure of the effect of buffer type on the protease but it might
be possible that the proteinase K is the problem.
My experience is that they can vary between lots.

I would suggest checking the fixation. We had similar problems if the
tissue was not completely fixed.
Are the tissues free to move around in the cassettes (ie have free
unhindered access to the fixative)?

totally neutralized and you will need to add more ammonia.

This will affect the haematoxylin staining.
To improve the staining, try pretreating the dewaxed sections with 1%
periodic acid for 10 minutes prior to HE staining. This might work.
