[Histonet] Influenza A IHC

If anyone is still doing Influenza A IHC testing, could you share your
vendor source and catalog number with me?  I'm looking to find a better
source than what I'm using right now.  I don't do it very often (since we
have a PCR test) but once in awhile it's requested, and my old antibody
just doesn't seem to detect all that it should.

Hi Sandra,

Being a mouse monoclonal, it may not cross-react with canines/felines; the
epitope may be specific to human follicular cells only.

The polyclonal Thyroglobulin from Dako/Agilent works fine on all the
mammals I've tried it on so far (and a frog as well).

We cut CD31, CD45, CD45RA, CD117, CD204, and MUM1 controls fresh (same
morning as staining; case material as well).

We've never had any problems with Synaptophysin; it's always worked very
well on unstained controls that are up to a year old (stored in the fridge).

Hello,
It depends upon which 'red' chromogen you are using.  There are several
with different chemical compositions, so you will need to look at the
Safety Data Sheets which pertain to the chemical you are using.

In my lab we use 3-amino-9-ethylcarbazole (AEC) which is considered
hazardous.  It causes eye and skin irritation, may cause serious eye
damage, and may cause cancer.  We ship our waste off for proper disposal;
it does not go down the water drain.

[Histonet] guinea pig IHC

Has anyone ever performed IHC on frozen sections of guinea pig tissue?  I
am experiencing an enormous amount of bubbling when doing the peroxidase
blocking step, even though I'm only using a 0.3% concentration of H2O2.
And when I say 'enormous', I mean it's like continuous champagne bubbles
rising out of the tissue, even after 20 minutes in the H2O2 solution.

I can't find anything in the literature that mentions guinea pigs having a
higher peroxidase content in their tissues.

Hello Paula,

This isn't an answer to your question, because I do use the Millipore
antibody (MAB805) and it works quite well on a Dako platform or done
manually.  It requires enzyme digestion antigen retrieval.  I use it at a
1:400 dilution (45 min) with a polymer detection system (45 min).

I'm not sure why it works on one platform but not another.

Hi all,

I do have MAC387 and it works great in dog and pig tissue.  The PI is
asking for additional CD markers to macrophages (CD68 and 204).  I need
them to work on dogs and pigs, but of course, there's not many vendors who
list species cross-reactivities on their data sheets beyond the usual human
and mouse.

In my earlier trials years ago with CD68 (clone KP1), I didn't have success
using it on dog tissue.  Am looking for a different clone that will
cross-react with dogs.

When selecting infectious agent positive controls, I also select cases
which have been shown to be positive by other testing means (EM,
Bacteriology, Virology, PCR).

In addition, I run a cross-reactivity validation with my antibody on other
infectious agent positive control slides to make sure my antibody is
specific for the infectious agent and doesn't cross-react with other
viruses/bacteria, etc.  Results go into a table that is stored along with
the initial antibody validation/optimization logsheet(s).

When validating tissue antigens/tumor markers, I use the obvious normal
anatomic cell/tissue control for that antigen.  My anatomic pathologist
advisor signs off on the final optimization for all antibodies before the
IHC test can be put into public use.

[Histonet] Aperio slide scanner

We often do IHC testing for Rabies on formalin-fixed, paraffin-embedded
tissues here, with no extra precautions needed.

Dr. Raff,

You will need to do trial runs to first determine if the antibody can
detect the specific epitope in animal tissues (epitopes aren't always
conserved between species), and then do your own optimization to determine
the best dilution and tissue pretreatment for that particular species.
 They can vary by species, and don't always coincide with the
recommendations on the antibody data sheet provided by the vendor.

To calibrate a pH meter, we use purchased pH buffers at pH 4.0, 7.0, and
10.0.

To perform the IHC method, we use TBS (pH 7.6) with Tween 20 as our rinse

I work in animal tissue only.  I also agree that an open autostainer system
works best, since you may have to run multiple dilutions and incubation
times for the same antibody because of the different species' reactivities.
 You may also need multiple types of reagents to cover detection in
different species, so you'll need a system which will allow you to use
multiple vendor products.

An aside... The CD4 and CD8 Abs that I've done on various animals are
usually only done on frozen sections, so I tend to perform that IHC
manually, in order to be able to rinse slides more gently than an
autostainer does (and do overnight incubation with primary Ab, if
necessary).

 Doing ihc on animal tissue is very tricky, if for no other reason than you
 are trying to standardize an antibody across multiple species. You will
 need to use every trick in the book and invent some of your own to get
 reproducible results.

 I would steer clear of any platforms that do not let you customize
 everything. I would suggest an OLD Dako or biocare's IntelliPath. Both have
 their quirks but they have the openness you require.

I've had it to work in cat, cow, dog, fox, horse, otter, pig, and raccoon
in my lab.

According to vendor - it's supposed to also work in guinea pig, monkey,
rabbit, and rat, but I have not tried it in those animals

Needs either enzyme digestion or heat antigen retrieval.

According to our Receiving department, each country/institution has its own
permit requirements for receiving samples from outside of their own
country.  It is up to the shipper to inquire of the receiver as to what
permits are needed to ship to that country (Germany, in your case).  My
source also said that FedEx would also have good information on this, since
they ship nearly everywhere around the globe.

  I have been aware of some edge effect on an occasional slide, but
 lately we noticed the effect is showing up in the area of the slide that is
 farthest from the labels. We use DAKO Link Autostainer and their
 pretreatment baths for everything. One case in particular we stained a
 large melanoma met to colon that was well fixed and processed that stained
 only the edge, however when we sent block to Neogenomics reference lab,
 their slide stained evenly over the entire tumor area. We both used a
 melanoma cocktail. Not sure what is going on. Any suggestions?

Patti,

You're using a mouse monoclonal antibody on mouse tissue, so be aware that
the secondary link antibody will bind to any mouse IgG it finds in the
tissue (normal plasma cells, IgG in normal sera present inside blood
vessels, or as infiltrating interstitial normal sera, etc.), not just to
the primary anti-Actin antibody.  You may have to use a mouse-on-mouse kit
for the Actin staining portion, if you see too much nonspecific background
in your tissue.

 BioReliance sells a good antibody (clone 7H2), in case anyone else out
there wants to know a vendor for the antibody.  Enzyme digestion works OK
for antigen retrieval, but HIER works much better at exposing epitopes.

Clarification to my previous post - we can't *accept* human tissue here.
 I'm sure the antibody will work on any species, however.  The antibody is
specific to the virus itself, not the tissue species it's residing in.

You need to do it sometime before the HRP-labeled conjugate is put on.  If
you put it on after the HRP conjugate, you'll block the HRP substrate and
won't see where the primary antibody is binding.  I usually do mine before
antigen retrieval, but have done it after the primary antibody step on
frozen sections and cell smears.

Hi Margaret,

We have used CK-Lu5 in the past with great results.  It stains dog, cat,
pig, cow, horse, goat, deer, bird, frog, hedgehog, snake, guinea pig, rat,
and bats so far.  Use Proteinase digestion for antigen retrieval.

We did notice that in recent times the staining intensity diminished with
CK-Lu5 on some runs and we had to keep dropping the dilution down to more
concentrated levels to achieve the same staining intensity as in the past.

Dako's CK-MNF116 is also a great broad spectrum (also stains pretty much
the same animals strongly).  However, CK-Lu5 stains more hepatocytes than
CK-MNF116 does, in my experience.

I've had 3 Dakos and have had the drying phenomenon in all of them to some
extent.  We put wet paper towels in the stain sink, as others have
mentioned.  Plus, we found that most of the problem came from having an air
gap between the hood and the unit, which wasn't closed entirely by the
gasket.  (The circulating room air got through the gap and dried out the
slides in that front row area.)  We use a long piece of styrofoam padding,
cut it to length, and insert that between the bottom of the closed hood and
the metal base at the beginning of each staining run, and it blocks all
unwanted airflow into the slide chamber.

NY in winter has low humidity, I still have no problems with my Dako
stainer. Make sure the lid is closed. I had a tech who consistently left the
lid open, there was some drying on the first row of slides. Closed the lid
the problem stopped. Are you using Dako buffer? The surfactant in the buffer
should prevent drying. You might want to run it by the Dako tech service.

Renee, it can depend on where you are: Florida? 70, 80% humidity, no drying
out. South Dakota, in winter? 10 percent humidity and you get drying
problems.

A good auto stainer (like DAKO) with adequate amounts of dispensed reagents
during the correct periods of time should not experiment any drying out on
the slides. Adequate humidity is required to be controlled during manual
IHC, especially if done over a heated support. If because of any reason
(including not leveled slides) you experiment drying out, the best way would
be to have an open flat dish containing water but, again, that was never a
problem for me using the DAKO auto stainer.

We are wondering what other labs are doing to control the humidity while IHC

Our policy is that if a radio station is to be played in the lab, the
choice has to be a unanimous one of everyone working in the lab, it cannot
be talk radio (prone to political leanings), it has to play appropriate
music for work setting, and it has to be turned down low enough to almost
not be audible.  Personal iPods with only ONE earbud used is preferable to
a room radio station.

I personally would not use it; you don't know how much loss of immunologic 
reactivity has occurred, and that might affect your project data.  Buy a

on human tissue and kept it in -20 for a while. It came in liquid but now 
it's all stiff

and looks like it's being crystallized. I cannot pipet any of it. It
has hardened and looks like a piece of glass at bottom of its container. I 
tried to re-suspend it with the same amount of glycerol, but the

antibody doesn't seem to be mixing with it.

Does anyone know how to save this antibody? How can I bring it back to its 
liquid state?

48 hours, and then frozen?

current lot at around 1:5000 with HIER (Decloaking Chamber).  If you use 
another method of antigen retrieval, drop the dilution down quite a bit.

store the slides (fresh frozen unfixed) at -80 in freezer bags, but they

CD68 - clone KP1 has not worked on sheep (also not dogs, cows, horses, and 
chickens) in my experience.  I have found it to stain primates well (and 
rare cells in pigs and cats).

When taking coursework eons ago, one of my professors addressed this very 
issue.  He said that '-ic' and '-al' were BOTH adjective suffixes... and 
there was no need to include both at the end of a noun to turn it into an 
adjective (redundancy).


So 'anatomic' it is.  And it follows that another correct word spelling is 
'pathologic', not 'pathological' though the latter is commonly used.

If you have no luck with calling your vendors or finding an MSDS link on 
their website, simply Google your chemical name and the letters MSDS after 
it (such as: sodium chloride MSDS).  Up will pop a long list of companies 
who have them available.  Find the format you like the best and print out 
for your files.  This should do, in lieu of one from the actual vendor, as 
long as the CAS number is the same as you use in the lab.

We have successfully stained ferret tissue using an antibody to Feline 
Infectious Peritonitis Virus (which also cross-reacts with swine 
Transmissible Gastroenteritis Virus).  We buy our antibody from Custom 
Monoclonals, Inc. and use heat-induced epitope retrieval (pressure cooker 
and target retrieval solution).  If you would like more details than this, I 
can send an uncontrolled copy of our SOP to you.

IMHO: Running any piece of tissue as a control that does not belong to the 
patient being tested makes zero sense.  Because it would not be from the 
patient tissue being tested, how do you know if it was handled the same as 
the patient tissue?  For example:


1) Were they processed the same way?
2) Did the patient tissue dry out in the OR before it was delievered?
3) Was the patient tissue ever irradiated?
4) Does the patient tissue contain any of a number of substances that could 
cause non-specific staining.

5) Was the patient abducted by aliens?

My point is that running a piece of tissue as a negative control that is not 
even from the patient being tested throws all of the conditions that the 
patient tissue was exposed to prior to and during processing out the window. 
This makes NO sense.

I received slides on sentinel lymph node biopsies with a positive
control
on the same slide as the breast tissue, but the negative control was

the patient's lymph node and did not have the corresponding section used

the positive control.  The patient's own tissue cannot be used as a
negative
control.  The tissue that stained positively must serve as the negative
control without the antibody.  This is critical and you need to correct
that
immediately.

[Histonet] esterase stain

Fw: [Histonet] CD79 alpha dilemma - ANSWER CORRECTION

Hello everyone - my apologies for posting a partially incorrect answer last 
week.  I was looking at the wrong line on a spreadsheet for CD79a abbreviated 
protocol information and wrote down the wrong HIER information.

What we do here currently with HIER for Biocare's CD79a (clone HM47/A9) on 
dogs, cats, pigs, cows, etc., is unmask antigens using:  0.1M Citrate buffer, 
pH 6.0 in a Biocare Decloaking Chamber.  (High pH retrieval solution does 
expose more antigenic sites, but it damages the tissue more when using the 
pressure cooker method.)  In the past we used high pH retrieval solution when 
doing HIER using a microwave oven (gentler method).

Hi Craig,

I have found CD79a from Biocare (cat. # CM067C) to stain dog, cat, pig, cow, 
horse, sheep, and mouse so far.  Have not tried it on other species yet (in 
other words, I don't have any negative species results to report).


I do have to use HIER with high pH retrieval buffer (Dako; contains EDTA) to 
expose antigens (in a Biocare Decloaking Chamber/pressure cooker).  Citrate 
buffer doesn't do the job well enough.  Microwave oven antigen retrieval 
will also work, but it exposes fewer epitopes than the pressure cooker.

the block, it appears white and crumbly.  The funny thing is some blocks

out all right, but most didn't.

PLEASE help!  Let me know how you process your animal fat (sp. Pig)! Is
there a
size issue (we trim it if it is greater than 5mm)?  We have gotten help
from the
histonet before and instituted these suggestions (i.e. let sit in
formalin for
48 hours; put in cassettes in a 60 degree oven for 1-2 hours and then
cool to
room temp. Then process using a 1 hour program).

Any help would be appreciated.

I know that Ki-67 from ThermoScientific (cat# RB-9043) works on bovine 
tissue, in my experience.  It does not stain a large quantity of cells in 
bovine tonsil, but those that do stain positively stain with moderate

do just DAB without blocking with h202 for myleoperoxidase but it will not
distinguish between eosinophils and neutrophils myeloperoxidase will stain

So where the microtome is here that I have to use we have to wear those
blue hospital booties and disposable lab coats (the white paper type
ones).  With me and several other people walking around in those booties
the amount of static electricity is to say the least frusterating!!

To clarify even further:  we cut 2 patient sections, put one on a slide with 
a section of positive control already on it.  This slide gets stained with 
the antibody  The other section goes on another slide and is run as a 
corresponding negative control using the same antibody protocol but 
substituting a negative control serum for the antibody, thus this is a 
negative reagent control slide.  Elements within the patient slide that 
received antibody and are expected to be negative, serve as a negative 
tissue control.  Again, we run 1 negative control slide for EVERY 1 patient 
block in a run but only 1 negative control per any number of antibodies run 
on that same block, using the harshest protocol.  Only autopsy cases differ 
in that we run 1 negative control per TISSUE TYPE.

So for every HP you do, you process a control cassette with the patient
tissue cassette?  That seems like alot?  How do you get that many
control tissues on a daily basis?  What do you do with the remaining
tissue in the control block?  If you throw them away everyday, I would
be interested in some of them.  How do you know what IHC stains the
pathologist is going to order to know what control tissue to fix and
process at the exact same time?  We have always just had a bunch of
blocks that you cut a control from?  I understand that there is
variability with processing, age, etc. not trying to be dense just still
don't understand... Most places I have ever worked have control blocks
that they cut a fresh control from everyday, then stain with the patient
tissue.  If there are 3 HP cases, from what I am understanding, you guys
are saying you need 3 controls for slides that are on the same machine,
with the same reagents, same antibody, and same times.  Why couldn't you
just have one for all 3 cases?  Then the next day have a fresh ONE for
that day, date them, and file them.  So if you needed to see the HP
control for October 15th, you could go pull the control for that day...

Because each tissue block has its own characteristics regarding fixation
and processing some of which can influence the reactivity. If you have a
bank of negative controls, how can you be sure that any of those blocks
have received exactly the same treatment and reacted in the same way to
the test block?
The same goes for any bank of positives, so that is why you should have
a positive control section in the same slide as the test section.

Mala,

If your main problem is that the reagents aren't spreading across your 
slides, I agree that you should put Tween 20 (0.05% per volume) in all of 
your rinse buffers between steps, regardless of if you do your IHC stains 
manually or with an autostainer.


If this doesn't solve the problem, post your method on histonet so that we 
know more about your procedure steps and can send additional advice.

Our pathologists have made a complain that the staining of slides have
been a problem.  Same antibodies which stain one day does not work the
following day.  This has been going on for a few months.  When we repeat
stain they either work or not work and we have been sending them to
reference labs.

Our tech support person was here and told us that the slides ( fisher plus
slides) have been a problem with some of her customers.  Meanwhile our
surrounding labs which are using the same slides have no issues.  She
showed us how the reagents were not spreading properly on the slides.

Is anyone confronted this issue?  If so,  how was it solved?

Retrieval solution (Dako)
Primary antibody - anti-West Nile Virus (BioReliance, cat. # 81-002; clone 7H2; 
working dilution at 3-6 ug/ml Ab concentration, depending upon lot)
Secondary linking antibody - EnVision+/HRP goat anti-mouse IgG (Dako; with 2-4% 
normal case species serum added per volume)

Reminder... these 'please send money' or 'please send your bank acct 
information so that I can deposit money into your acct' pleas are ALWAYS 
scams.  They have been for years.  If a trusted 'friend' was truly stranded 
in a foreign country, they'd borrow a phone and call home.  If a long lost 
relative died and left you money in another country, it would be a lawyer 
contacting you via telephone or letter, and definitely not through an email 
message.


Best advice... don't even open these messages up.  They're bogus, and could 
be carrying viruses, worms, etc. themselves.  Being on a listserv like 
histonet, we're prone to receive a lot of these email fraud attempts. 
Ignore them.

The email you received is a Nigerian Scam.  Someone was able to  hack 
into Jerry's computer and get all his contact list!  You  happened to be

October.  Fortunately for me, I didn't have  any bank account 
information in my computer.  I have a MAC and so  I went to the Apple 
Genius Bar and they checked my computer for  any viruses.  Fortunately, 
it is very difficult to get into a  MAC.  This has been on the news in 
Idaho and various other  states.  DO NOT RESPOND to any emails that want 
any personal  information!!!  You need to clean-up your computer from 
any viruses.

vomiting and wasting disease and/or motor disorders (incoordination,

Please help, we have had an increase in demands for repeats on our AFB 
stain. Our pathologists have been noticing what they believe are false 
positives on our slides. We perform our AFBs on the Ventana Nexus and some 
are done by hand. Because of this problem, we have been doing side by side 
comparisons using the Nexus and hand stain.
We still have complaints. We have made new reagents, changed kits, tested 
our DI water sytem and have different techs cutting using different 
waterbaths.

Any advice or comments would be greatly appreciated.

I work on all species, porcine being one of them.  I've never done a % 
survey of the number of Abs made against human epitopes that also work on 
porcine tissue, but I can give you generalizations.


All of the rabbit polyclonal antibodies that I've tried have cross-reacted 
with porcine tissue.  The polyclonals that I use the most are the Abs 
against hormones, neuroendocrine markers, vasculature, astrocytes, etc.
Most of the mouse monoclonal antibodies that I've tried have cross-reacted 
with porcine tissue.  These would be the intermediate filaments, melanoma 
antigens, cell proliferating antigens, neuroendocrine markers, etc.
You may have more difficulty with leukocyte markers (CD markers).  They seem 
to have less homology with human markers than the above items, and not all 
cross-react, in my experience.  You only learn this by trial and error, 
unfortunately, since data about species cross-reactivity is rare.  I have 2 
polyclonal and 2 monoclonal CD markers that cross-react with porcine.  That 
is not to say that there aren't more, but that's all that I currently have 
worked up on porcine tissue.

I have not tried Abs like ER, PR, CEA, TTF, etc.
I've done species cross-reactivity studies with every antibody I have in 
stock (80), so if you send me a list of the antibodies you're interested in 
(directly to me), I can let you know which ones cross-react with porcine 
tissue, and which vendor works for me.

I work with some porcine tissues and it is sometimes difficult to find 
antibodies specific to pig. A doctor I work with asked some one he knew in 
Europe about using antibodies that are listed as working in human being used 
on porcine tissue. This person stated that there would be about a 10% chance 
that the anti-human antibodies would work on porcine tissues. The antibody 
companies mostly give the same we did not try it on pig tissue response. I 
understand that it is likely expensive to continue to test antibodies on a 
wide variety of animal tissues, so that is fine. But what do you guys think 
about the 10% chance? I would have said more than that.

Lynn,

I don't know anything about the Ventana Benchmark XT requirements, but I 
have never had any problem with Vimentin on a Dako Autostainer and also done 
manually on all sorts of animal tissue - with the exception of mouse.  The 
V9 clone doesn't work on mouse, in my experience.  (It'll work on frogs, 
however; go figure.)


My protocol uses Dako Target Retrieval in a Biocare Decloaking Chamber for 
antigen retrieval, 45 min in primary antibody, 30 min in EnVision+/HRP goat 
anti-mouse IgG, and 10 minutes in AEC.  Very strong staining.

[Histonet] veterinary IHC question - BVD Ab

Hi all,

I've been using the BVD clone 15C5 for years, but need to find another vendor 
source with reactivity to another clone.  Does anyone have experience with and 
recommendations for a commercial source of a monoclonal BVD antibody that has 
strong reactivity?  This would be for Type 2 BVD virus.

I pack slides in 100-slide plastic boxes, with gauze sponges laid inside the 
boxes on top of slide edges to ensure that they don't jiggle around inside 
the box.  Boxes are taped shut, then packed inside a cardboard box with 
plenty of room for packing peanuts or bubble wrap.  Never have had a 
problem.

[Histonet] veterinary lab question - chem expiration

What are veterinary labs (non-CAP inspected labs) setting as their expiration 
dates for dry and liquid chemicals that come without expiration dates on the 
labels?

On the Histonet I've found varying answers from human labs (5-10 yrs for dry; 
1-5 yrs for liquid).  I'm in the process of setting guidelines for my labs, but 
want to be sure to be in agreement with other vet labs out there as well.

I don't use the ready-to-use GFAP from Dako, but I do use the concentrated 
form of GFAP (it is also a rabbit anti-cow GFAP.)  It's worked successfully 
in dog, cat, pig, cow, horse, goat, sheep, deer, eagle, mouse, rat, gerbil, 
guinea pig, hamster, monkey, ferret, and kangaroo with no background 
problems whatsoever.


The EnVision+HRP Rabbit will only bind to the rabbit IgG, not the cow IgG, 
so that is not the source of your background.  I would suspect that if 
you're getting brown staining on the slide itself, your antibody is much 
much too concentrated.  My working concentration calculates out to be 4 
ug/ml of Ab.  You might see if your ready-to-use preparation has a 
concentration listed, and then work from there to approximate a 4 ug/ml 
concentration with diluting it out.


If that doesn't work... buy the concentrated stock form and optimize it for 
your tissues.  In my lab, it requires absolutely no tissue pretreatment, and 
the stain is always strong, clear and clean.  Good Luck!

We are using DAKO's ready-to-use GFAP antibody on our bovine tissue and 
using Envision+ HRP Rabbit detection system.  When using this antibody we 
see a brown color on the slide itself.  The staining was intense and quite 
uneven.  We have diluted the anitbody to 1:10 but there is still the brown 
slide.  The staining is not quite as uneven but appears more washed out. 
We don't see this with any of our other antibodies we are using and our no 
primary controls for GFAP are clean.  Any advice or explanations for the 
slides picking up the brown color or improving GFAP IHC on bovine tissue?

Dako's Factor VIII (von Willebrand Factor), cat. #A0082 works for me on 
canine tissue (and cat, pig, cow, horse, sheep, chicken, mouse, etc.).  I 
use Proteinase K enzyme digestion pretreatment for 5' at room temperature; 
primary rabbit Ab incubation 30', EnVision+/HRP anti-Rabbit 30' (with 4% 
normal dog serum added per volume for blocking), AEC 10'.

[Histonet] AAVLD Histo sample block slide retention policy

For veterinary diagnostic lab personnel:

We're having a difference of opinion here over how long our Histo slides need 
to be retained.  It was my understanding that all paraffin blocks and stained 
slides need to be retained for 7 years.  Others here believe that as long as 
the blocks are retained, it's not necessary to retain the stained slides past a 
year or two.  Storage space might be a problem in the near future.

I've looked through the AAVLD Essential Requirements, but cannot find a 
definitive answer to this question.  Can someone help me out with the correct 
answer?  We're writing a sample storage SOP for each lab section today, and I'd 
like to get an answer as soon as possible.
