Yes, it does. Currently in the newer powerbooks, you have essentially two speeds--4200 rpm and 5400 rpm. Obviously, a faster drive will access files more quickly. A larger drive if it's fragmented badly may take longer to access a file. I'm sure Remy of Insanely-great-mac.com has some benchmarks.
