Well, you need to put the drives where a.) You need security and/or b.) you have a
particular directory that's gonna hold a LOT more (like /home). I can't tell you
where these directories are; it depends on what you do.
I wouldn't bother with one on /opt, since I've seen such a small number of apts
(possibly KDE) that still use that odd naming scheme. /tmp is a good one, and
possibly others, but -especially in machines with lots of users- /home is a good thing
to go onto another drive. Heck, it's also a good candidate for RAIS, even if it's
just software raid; it can speed up the access, and as long as you use two seperate
drives on seperate buses, it gives you more reliabilty...
